Development of Live Food Culture for Brackishwater Fish and Shrimp Specis

Abstract/ Executive Summary:

Maximum growth of Chlorella sp. was observed at the 7th day of culture period in all culture media. Highest cell growth (302x104 cells/ml) of Chlorella sp. was found in modified f/2 media. Maximum growth of Chlorella sp. in BFRI medium and Dix mix was 272x104 cells/ml and 232x104cells/ml, respectively. Microscopic observation of the water revealed dominance of rotifers (Brachionus plicatilis), which were then picked up using micropipette. Purification of Brachionus plicatilis was done in the laboratory through several culture and segregation in succession system. After eight days of culture, highest population of rotifer (45.67 nos/ml) and N. oculata (43.44 nos/ml). The difference in growth of rotifers fed with different algae was significant (p<0.05). Final number of egg bearing rotifer fed with Chlorella sp. was highest (27.76%) and significantly different (p<0.05) from those of rotifers fed with N. oculata (24.60%) and T. chui (15.77%).

Objectives:

To develop the most suitable methods of live food culture for brackishwater of marine hatcheries in Bangladesh so that the culture of a wider variety of target culture species may be possible in future. The specific of the program are : Mass culture (indoors) of Marine Chlorella sp.; Isolate local microalgae (Chaetoceros sp., and Skelotonema sp.) and zooplankton, rotifer (Brachionus plicatilis) food species from ponds of BS pond complex, nearby rivers and gher areas; and develop protocols to maintain pure stock culture of isolated microalgae and zooplankton in Laboratory.

Methodology:

Chlorella sp. Was segregated from the mixed phytoplankton, collected from local brackishwater environment. The segregated Chlorella sp. Was purified through successive culture and segregation system. Comparative efficacy of three culture media viz., BFRI media, Dix mix and modified f/2 media was tested in cylinder fibre glass tank (200L0 for the mass culture of Chlorella sp. For this purpose, 14 ppt underground brackishwater was filtered through 1µ filter bag to remove particulate matter. Than this water was chlorinated with bleaching powder. Aftet 30 minutes, the water was declorinated with aeration and sodium thiosulphate @0.175 g/L treatment. Pure strain of stock Chlorella sp. was cultured in 20L carboys and inoculated in the tanks at the concentration of 120x104 cells/ml. Temperature of water in the tanks was maintained at about 25±1⁰C. The culture was continued for 10 days. Growth of algal cells was estimated daily through enumeration with haemacytometer.

Achievement/Findings:

The project will assist in paving the way for the hatchery culture of species such as Scylla serrata and Lates calcarifer and other suitable finfish/crustaceans. Once the protocols have been developed for the culture of the appropriate live food species for each of the larget culture species, the changes of success will increase. The collection of or isolation of local species of live food organisms and development of protocols to culture them efficiently will be of benefit to nutrition of the local target species in culture.
