Genetic Variability Among Local Population of Graspid Crab, Scylla Serrata and Freshwater Giant Prawn, Macrobrachium Rosenbergii

Abstract/ Executive Summary:

The food of mud crabs comprised mainly of crustacean animal (45-50%), followed by gastropods (15-40%), small fishes (10-20%), unidentified debris (5-15%) and plant materials (2%). Zooplanktons (Copepods) were occasionally found in the stomach of mud crabs. The smaller size crabs prefer plant materials, while bigger crabs prefer meat as their feed. Highest survival, 23.3% was found in 25 ppt and the lowest, 13.3% was in 5 ppt saline water. The highest specific growth rate of crab (2.8% /day) was in 20 ppt and lowest (2.77%/day) in 10 ppt saline water. The sub-adult mud crabs was found to ingest plant materials, but the adult mud crab, prefers animal materials (snail/gastropods, crustacea, fish etc.) as their food. Salinity level ranging from 10-25 ppt may be suitable for culturing of crablings, but the higher salinity was found little better for the survival of mud crab.

Objectives:

To provide an overview of genetic structure of Scylla serrate and Macrobrachium rosenbergii; to understand genetic variation among local population of Scylla serrate and Macrobrachium rosenbergii; and to estimate the genetic distance among the population of S. serrate and M. rosenbergii.

Methodology:

Horizontal starch gel electrophoresis was used in order to estimate the degree of genetic differentiation. Samples of crab and prawn were collected from south-west coastal region and were kept at -20⁰C prior to electrophoresis. The tissue to be analysed for allozyme was homogenized with approximately of equal volumes of the fragments of frozen muscle and distilled water. Then the homogenates were centrifuged at 12,000 rotation per minutes for 12 minutes at 4⁰C and supernatant absorbed by filter paper and used for electrophoresis. Horizontal starch gel electrophoretic techniques, buffer systems and staining procedure was followed by the method of the Japan Fisheries Resource Conservation Association.

The main purpose of this study is to find out the genetic distance among the local samples as well as to know either there is only one/more species of Scylla or Macrobrachium in Bangladesh or not.
