Standardization of Inoculation Methods of Sclerotinia Sclerotiorum in Mustard

Abstract/ Executive Summary:

Five inoculation methods of Sclerotinia sclerotiorum were tested to mustard plant. Of them Toothpick method showed best performance for establishment of the pathogen followed by Mycelial disc method. The rest three methods failed to establish.

Objectives:

To develop a easy and suitable inoculation technique which may be exploited for varietal screening of different crops against S. sclerotiorum

Methodology:

Seeds of BARI-Sharisha-9 were sown on pot soil (15 cm diameter). After germination only four plants were allowed. The fungus Sclerotinia sclerotiorum was collected from mustard plant, isolated and purified on PDA. The following methods of inoculation was demonstrated: i) Mycelial disc ii) Soil inoculation with mycelial suspension iii) Spraying of mycelial suspension on plant iv) Soil inoculation with sclerotia v) Toothpick method In case of mycelial disc method 2 mycelial disc of 7 mm diameter cut from the margin of 5 days old culture on PDA and placed at the middle portion of main stem. Inoculated portion was covered with moistened cotton and wrapped with plastic tape to ensure moisture. In case of mycelium suspension method 30 ml of mycelial suspension was drenched at each plant base (soil inoculation) while same amount of mycelium suspension was sprayed on whole plants with hand sprayer. Regarding sclerotia inoculation 30 sclerotia per plant was added in soil around the plant base. In toothpick method the fungus was multiplied on potato broth having with sterilized toothpick. Colonized toothpick was inserted at the middle portion of the stem. The inoculated portion was covered with pre-soaked cotton layer and wrapped with plastic tape to conserve moisture. Regardless inoculation methods all the inoculated plants were covered with poly bags for 48 hours. The optimum soil moisture at the base of the plant was maintained throughout the experiment. In each method of inoculation, 5 plants for each was maintained.

Achievement/Findings:

Out of five inoculation methods tested, two methods namely, Mycelial disc and Toothpick responded excellent for establishment of the fungus, S. sclerotiorum. They showed 100% infection. Between them toothpick required only four days for symptom expression and 8-10 days for total death of plants while corresponding values in Mycelial disc were 7 and 14-20 days (Table 10). The rest methods Mycelial suspension spray on soil and plant, and soil inoculation with sclerotia became ineffective.
