Diversity of Pathogens Causing Rhizome Rot of Ginger

Abstract/ Executive Summary:

An experiment was conducted during 2010-2011 cropping season in Regional Agricultural Research Station, Jamalpur to know the diversity of pathogen causing Rhizome rot of ginger in Jamalpur region. Fifty rhizome samples were randomly collected from 10 locations of Jamalpur, Sherpur and Tangail district. Among 50 samples, 41 samples were found diseased and 9 were found disease free. Among the 41 diseased samples 85.4% was found infected with Sclerotium, 51.2% was Pythium sp and 12.2% was Fusarium sp. The growth of the sclerotium was very fast and Pythium takes time to form pathogenic structure.

Objectives:

To identify the organisms associated with rhizome rot of ginger in Bangladesh.

Methodology:

The experiment was conducted during 2010-2011 cropping season at Regional Agricultural Research Station, Jamalpur. During the survey of disease incidence, a total 50 samples (Diseased and disease free) mostly deshi ada were collected at random from 10 ginger growing fields of Jamalpur shadar, Nandina, Melandah, Sreebordi(Sherpur) and Kushumhati Sherpur and local markets of Jamalpur and Sherpur. Samples of rhizome and pseudostem were collected from farmer’s field and only rhizome was collected from the local markets. Samples were taken in a poly bag then it is inserted within a brown paper bag. Before pathogen isolation the samples were kept in the refrigerator for a while. Isolation of pathogen Individual infected specimens of ginger, viz. pseudostem and rhizome were washed and cut into small pieces of 1.0-1.5 cm2. These pieces ware surface disinfected with Clorox solution for 1 minute. Four pieces of pseudostem and rhizomes from each samples were placed in petriplates containing PDA. Then the plates were incubated for 25.4^oC at 72 hours. The hyphal tips of fungi growing from these pieces were transferred to new PDA plate for pure culture and maintenance. After pure culture the growing fungus were observed under compound microscope for proper diagnosis of the fungus.

From the study it was revealed that out of 50 samples 41 samples were found diseased and 9 were found disease free.
