Effect of Trichoderma Against Seedling Blight Disease of Wheat Caused by Sclerotium Rolfsii

Abstract/ Executive Summary:

The formulated biological control agent Trichoderma harzianum was mass cultured in compost materials and tested against seedling blight disease of wheat caused by soil-borne pathogen Sclerotium rolsfii. The spore suspension of T. harzianum was also used as a seed treating agent. It was observed that soil amendment with compost materials containing biological control agent T. harzianum was found as the best treatment for reducing seedling mortality, increasing plant growth as well as yield of wheat. Seed treatment with fungicide and Trichoderma spores suspension gave similar effect for decreasing seedling mortality and increasing plant growth and yield of wheat.

Objectives:

To observe the efficiency on multiplication of Trichoderma in composting media and to control S. rolfsii of wheat.

Methodology:

The experiments were conducted in the field, Plant Pathology Division, Bangladesh Agricultural Research Institute, Joydebpur, Gazipur. There were 6 treatments viz. (i) Control, (ii) Seed treatment with Provax, (iii) Trichoderma spore suspension-1, (iv) Trichoderma spore suspension-2; (v) Soil amendment with Tricho-compost-1 and (vi) Tricho-compost-2. The unit plot size was 2.5 m x 3m. RCB design was followed with 3 replications. T. harzianum was multiplied in the mixture of khesari and wheat bran with mustard oilcake as main substrates. The formulated Trichoderma was used to mass multiplication in the cow dung based composting materials. Two different cow dung based compost materials were used which called Trico-compost-1 and Trico-compost-2. T. harzianum were cultured in PDA and PBD media and spores were harvested from 10 day-old cultured used for seed treatments. Soil was inoculated with S. rolfsii colonized mixture of khesari and wheat bran along with mustard oilcake substrates @100g/m2 soil before seed sowing. The S. rolfsii inoculated soil was challenged with Tricho-compost @ 3 t ha-1In case of chemical treatments, seeds were treated with Provax @ 2.5 g kg-1 seeds before sowing. Data were collected on seedling mortality, plant growth parameters such as shoot height, shoot weight, root length and root weight. The yield of wheat per unit area was also recorded. Data were analyzed by the computing MSTAT program following the standard statistical procedures. Duncun’s New Multiple Range Test (DMRT) was carried out to find out the treatment effects.

Achievement/Findings:

The highest seedling mortality 41.0 % was recorded in the control treatment. Significantly lower seedling mortality 11.67% and 12.67% was recorded in soil amended with Tricho-compost-1 and 2, respectively followed by seed treatments with Provax and Trichoderma spore suspensions by 15.0%, 18.67% and 20.67%, respectively. Soil amendments with Tricho-compost performed as the better treatment in increasing shoot height, shoot weight, root length and root weight which was significantly differed from other treatments as well. The highest yield of wheat by 4.37 t ha^-1 was recorded in Tricho-compost-2 treated plot followed by Tricho-compost-1, seed treatment with Provax and Trichoderma inocula-2 where yield was 4.31, 3.95 and 3.82 t ha^-1, respectively. The lowest yield was recorded in control plot 3.25 t/ha. Yield was increased 25.63% and 24.59% due to application of Tricho-compost-2 and 1 and it was 17.72% due seed treatment with Provax. Seed treatments with Trichoderma inocula-1 and 2 increased yield 14.02% and 14.92%, respectively compared to control. From this study it is observed that soil amendment with compost materials containing biological control agent T. harzianum is proved to be the best treatments for reducing seedling mortality, increasing plant growth and yield of wheat. Seed treatment with Provax showed similar effect as seed treatments with Trichoderma spore suspensions for reducing seedling mortality and increasing plant growth and yield of wheat compared to control treatment.
