Estimation of Chlorophyll of Mulberry Leaf Infected by Cercospora Moricola Cooke Leafspot Disease.

Abstract/ Executive Summary:

Disease development of plant causes an imbalance situation in the photosynthetic pigment (Chlorophyll) which effects the utilization of light energy by the plant in the leaf. Yellowing or chlorosis on leaf is a common symptom of disease and is associated with tissues surrounding a nectrotic area. The chlorophyll may be degraded to develop in color or yellowing of the infected leaf. This is a common symptom of mulberry leaf spot disease due to Ccr-cospora moricola. In case of leaf spot disease, chlorophyll content is degraded. Mahadevan and Sridhar (1982) reported that disease interfares with the biochemical events occurring in the diseased tissue and culminates in reduced photosynthesis and yield.

Objectives:

To destruction of chloroplast, the functional laminar surface is considerably reduced and affect the productivity of the plant.

Methodology:

Five disc of 0.567 cm" area were taken from five different areas of leaves of each treatment separately. Chlorophyll was extracted with 80% aqueous acetone. The suspensions were decanted into centrifuse tubes and centrifused for ten minutes at 4000 rpm. The clear solutions were decanted and made upto 10 ml with 80% acetone. The optical densities of these solutions were determined against 80% acetone using a spectrophotometer at 645 and 663 run.
