Effect of Storage Period, Types of Container and Genotypes on Moisture Content, Vigor and Germination of Soybean Seed

Abstract/ Executive Summary:

The study was conducted to assess the effect of storage period and types of container on moisture content, germination and vigor of soybean seeds. The experiment was carried out at Oilseed Research Centre, BARI, Gazipur, during the period of August 2008 to march 2009. The seed were stored for seven month from August to March and the storage containers used were tin, plastic, earthen pot and jute sack. Results revealed that the storage container influenced seed moisture content and germination throughout the storage period. Rise in seed moisture content of all genotypes showed inverse relationship with the magnitude of seed quality attributes. In jute sack and earthen container seed moisture rose sharply and it provided adverse effect upon seed quality and thus seed quality retarded sharply. After storage in jute sack and earthen container, significant decrease of germination percent (up to 40.67-7%) was observed in all genotype but no appreciable change was recorded in seeds stored in tin and plastic container.

Objectives:

To evaluate the effects of storage period, types of container and genotype on seed moisture content and germinability of soybean seed.

Methodology:

The experiment was conducted at the Oilseed Research Centre, BARI, Joydebpur, Gazipur-1701, during August 2008 to March 2009. Fresh seeds were collected from field office, ORC of Bangladesh Agricultural Research Institute. Two soybean varieties (Shohag and BARI soybean-5) and an advance line (Amber) were selected for the experiment. Seeds were stored under ambient condition (Temp. 20⁰C-27⁰C). The treatments comprised of 3 genotypes viz. Shohag, BARI soybean-5 and Amber, 4 Types of storage container viz. i. Tin container, ii. Plastic container, iii. Earthen container and iv. Jute sack. Storage period commenced on 01 August 2008 and data were taken in every 01 month interval on for different seed quality parameters observations. The experiment was conducted following Completely Randomized Design (CRD) with three replications. Calculation of vigour value: Vigour value was employed to identify of the viable seeds which was assessed by using the results of the above mentioned germination test according to Khandakar (1982), using the following formula: (a/1 + b/2 + c/3 + d/4) × 100 Vigour value (%) = S Where a, b, c and d are seed germinated after 1, 2, 3 and 4 days from the start of the germination test and S is the total numbers of seeds germinated. Determination of germination (%): Four hundred seeds were collected randomly from each containers and set for germination test in four glass petridishes having 100 seeds in each. Seeds were evenly placed on the top of 11 cm Whatman No.1 filter paper in each petridish. Filter papers were then kept moist by adding 5 ml distilled water. From 2nd days after setting germinated seeds were counted and recorded daily for next four days. Seeds with radicle extended up to 1 cm in length or more were considered to be germinated.

Achievement/Findings:

Storage container influenced seed moisture content and germination throughout the storage period. In jute sack and earthen container seed moisture content increase rapidly and it provided adverse effect on seed quality. After seven months of storage in jute sack and earthen container, significant decrease of germination percent (up to 40.67-7%) was observed in all genotype but no appreciable change was recorded in seeds stored in tin and plastic container.
