Performance of Explants and Media for Culture and Regeneration Ability on Rice Genotypes.

Abstract/ Executive Summary:

Objective of this study was to determine the suitable explants and media composition for culture and regeneration ability of different genotypes. Culturing and regeneration ability of cultivars is an important criteria for improving rice through tissue culture. All the explants showed high callus induction, the highest (86.81) being shown by immature embryo. Different explants showed 0 to 100% callus induction. Immature panicle of BRRI dhan28 and BRRI dhab32 didn’t produce any callus in K (modified B5) medium. BR5 demonstrated high callusing from all explants.

Objectives:

The objective of this experiment was to determine the suitable explants and media composition for culture and regeneration ability of different genotypes.

Methodology:

Culturing and regeneration ability of cultivars is an important criteria for improving rice through tissue culture. Materials having such potentials would be helpful to incorporate useful traits through different biotechnological tools. Callus induction and regeneration frequency of three explants (seed, immature embryo, and immature panicle) are evaluated by using MS and KA medium. The callus was observed within 15 to 30 days, 7 to 15 days and 3 to 10 days from seed, young panicle and immature embryo respectively.

Achievement/Findings:

Among the varieties, high callus induction was observed in BR5 and BRRI dhan28 (100% in both MS and K) followed by BRRI dhan29 (85.71% in MS and 90% in K) and Taipei-309 (98.57% in MS and 68.57% in K). Callus size varied from 0.1-.6 cm and doth compact callus except in K medium. Only BR5 produced compact callus in both the medium in higher frequency. Immature embryos of all test varieties regenerated in both the medium except BRRI dhan40 in MS media. The highest regeneration percentage was obtained in the MS (88.57%) and K (84.00%) media from BR5 with immature embryos as explants. The green plant production was highest in K medium irrespective of varieties. Among the explants immature embryo gave the highest average regeneration (34.57) in most of the cases. We have got more plant from immature embryo than other explants within a short time.
