Integration of Carriers for Effective Formulation of Trichoderma Harzianum Against Soil-borne Pathogens

Abstract/ Executive Summary:

Different substrates such as Khesari bran, Wheat bran, mustard oil cake and Rice husk and their integration were used for effective formulation of Trichoderma harzianum bio-pesticide. The formulated T. harzianum bio-pesticide was tested against seedling disease caused by Sclerotium rolfsii of tomato, eggplant, lady’s finger and Indian spinach. The formulation was added to Sclerotium rolfsii infested soil @ 200 g/m2 soil in seed bed. All the substrates formulated with Trichoderma had significant effect in increasing seed germination and decreasing seedling mortality as well as increasing growth of tomato, eggplant, lady’s finger and Indian spinach. Among the treatments, integration Khesari bran with mustard oilcake and Wheat bran was found better carrier for formulation of T. harzianum.

Objectives:

To find out the most cost-effective formulation and use of T. harzianum as bio-fungicides and its effectiveness against S. rolfsii

Methodology:

The experiments were conducted in the Plant Pathology Division, BARI, Joydebpur, Gazipur during 2010-2011. Sick bed of S. rolfsii was developed in the net house of Plant Pathology Division, BARI, Joydebpur, Gazipur. The pathogenic fungi, S. rolfsii was multiplied in Wheat bran + Khesari bran substrates. Soil was inoculated with the Wheat bran + Khesari bran substrates colonized with pathogenic fungi for established the pathogen in the soil. T. harzianum isolate was cultured in Potato Dextrose Agar (PDA) medium. Three different substrates viz. Wheat bran, Khesari bran and Rice husk and their integration viz. Wheat bran + Rice husk, Wheat bran + Saw dust, Khesari bran + Rice husk, Khesari bran + Saw dust, Khesari bran + Mustard oilcake and Rice husk + Saw dust were used in this study. The requisite amount of materials for each substrate were thoroughly mixed in a 1000 ml Erlenmeyer flask and autoclaved at 121⁰C for 15 minutes for sterilization. The sterilized substrates allowed to cool down and then inoculated with 5mm diameter mycelial discs of 5 days old T. harzianum culture. Inoculated flasks were incubated at room temperature (25±2 ⁰C). For even colonization, the flask containing inoculated mixture was shaken thoroughly by hand at every 3 days interval. After incubation for 15 days, the contents were taken out from the flasks; air dried and used as a T. harzianum bio-fungicides. The prepared T. harzianum bio-fungicides were used @ 200 g/m² in sick bed of S. rolfsii, thoroughly mixed with the soil and was incubated for 10 days with proper soil moisture. Only different substrates also used for comparison. Two hundred seeds of tomato, eggplant, lady’s finger and Indian spinach were sown in each treatment. Data were collected on percentage seed germination, seedling mortality; shoot length, shoot weight, root length and root weight.

Achievement/Findings:

Different substrates and its integration showed differential potentialities on seed germination and seedling mortality of tomato, eggplant, lady’s finger and Indian spinach. Among the substrates, Khesari bran inoculum gave higher seed germination followed Khesari + MOC, Wheat bran, Khesari + Rice husk. Seed germination was higher when formulated T. harzianum was used compared to control as well as when only substrates was for soil amendments. The minimum seedling mortality was recorded in the plots treated with formulated T. harzianum where as it was higher in the plots treated with different substrates as well as in the control plot. Significant effect of formulated T. harzianum in different substrates on the growth such as shoot length, shoot weight, root length and root weight of tomato, eggplant, lady’s finger and Indian spinach. Among the treatments, integration of Khesari bran with mustard oilcake showed better performance in increasing seed germination, reducing seedling mortality and increased plant growth of tomato, eggplant, lady’s finger and Indian spinach followed by Khesari bran, Khesari bran + Rice husk and wheat bran compared to soil amendment with different substrates and control treatments.
