Bio-assay of Different Concentration of Mahogany as Botanicals Against Fusarium, Bipolaris and Alternaria

Abstract/ Executive Summary:

A study on efficacy of Mahogany leaf, seed extract and oil was conducted at RARS Jessore, during the year 2010-2011. Five concentrations of Mahogany leaf, seed extract and oil was tested against Alternaria sp from tomato, Bipolaris sorokiniana from wheat, Fusarium oxysporum from chickpea and Alternaria sp from mustard. Radial growth reduction and % inhibition was the highest in 50% concentration of leaf and seed extract and 5ml/L Mahogany oil in all the pathogen 50% concentration of leaf extract could inhibitthe growth 61.40% of Alternaria sp and 50.60% of Fusarium oxysporum compared to control. On the other hand, seed extract could reduce 87.70% of Alternaria sp and 70.11% of Fusarium oxysporum over control. Oil extract also showed good inhibition 94.11, 92.35, 94.15 and 93.36% of Alternaria sp. from tomato, Bipolaris sorokiniana from wheat, Fusarium oxysporum from chickpea and Alternaria sp from mustard over control.

Objectives:

To evaluate the fungicidal activity of Mahogany leaf, seed extract and oil against few pathogens in laboratory

Methodology:

The experiment was carried out during the year 2010-2011 using CRD in the laboratory of plant Pathology Division in RARS Jessore. Four pathogen Fusarium oxysporum from chickpea, Bipolaris sorokiniana from wheat, Alternaria sp. from tomato and mustard were used. These pathogens were collected from the infected plant part, isolated by single spore isolation method under compound microscope, and obtained pure culture on PDA media. For stock solution 100 gm of fresh leaves were collected and 100 ml stock solutions (w/v) were prepared by blending with distilled water. Then appropriate quantity of stock solution were mixed to make 10, 20, 30, 40, 50% media with final volume of 100 ml in each flask. Control received 20 ml of distilled water to make final volume of growth media for 100 ml. In case of seeds same procedure maintained to prepare different concentration of seed extract in PDA media. In case of oil 1, 2, 3, 4, 5 ml oil was mixed with final volume of the growth medium to 100 ml. Actively growing mycellial disc of 5mm diameter of the pathogens were taken from 7 days old pure culture, then transferred to petri dishes containing the above prepared media and incubated in growth chamber. Data were taken when the control plate showed near about full growth of fungal mycellium. In case of Fusarium it was 4 days after incubation, in Alternaria sp and Bipolaris 7 days after incubation. All the data were subjected to analysis of variance followed by mean separation through LSD test using compute software MSTAT.

Achievement/Findings:

All the concentrations reduced mycellial growth significantly over control plate. Fifty percentage concentration of MLE showed highest radial growth reduction and inhibition then control of Fusarium oxysporum (3.10mm and61.40%) and Alternaria sp.(4.13mm and 50.60%). Seed extract at 50% concentration also showed the similar result which is 0.50 mm and 87.70% for Fusarium oxysporum and 2.37 mm, 70.16% for Alternaria sp. Mahogany oil of 5ml/L concentration gave significant inhibition of all pathogens over control. In case of Alternaria sp from tomato growth inhibition was 75.29% where it was 94.11% in rovral (2ml/L) compared to control. Similarly Bipolaris sorokiniana showed 84.35% growth inhibition in 5ml/l MO and 92.35% in tilt over control. 5ml/L concentration of MO showed 75.63% growth suppresson over control in Fusarium oxysporum from chickpea where it was 94.15% in bavistin and 70.39% in Alternaria sp from mustard and 93.36% by rovral.
