In Vitro Propagation of Potato (solanum Tuberosum L.)

Abstract/ Executive Summary:

The present investigation was conducted to develop a protocol for rapid plant regeneration in potato. The newly emerge sprout of six potato cultivar were used as explants. Among the different treatment the combination 2.0 mg/L of KIN and IAA showed best response to multiple shoot and root regeneration. The in vitro regeneration and multiplication potentiality was highest in the variety Granulla followed by Cardinal and Diamont.

Objectives:

1. To establish an efficient plant regeneration protocol in potato

2. To study the potentiality of different hormone on in vitro regeneration in potato

Methodology:

Sub title 1: In vitro regeneration potentiality of potato under different hormonal combination .The cultivars were as follows - Diamant, Cardinal, Granulla, Ultra, Dheera and Provinto. Disease-free healthy potato tuber was treated with 50 ppm GA₃ for sprouting. The sprouted buds were used as explants. The different hormonal concentration was treated as factor 1 and the different varieties of potato were used as factor 2. Sub title 2: In vitro tuberization in potato. The well establish potato cultivar viz. Diamant was used. These explants were cultured in MS solid medium (pH = 5.8) in test tube. Sub title 3: Off-season sprouting ability in potato. Three well establish cultivar viz. Diamant, cardinal and Granulla were used for this works. Five different concentration viz. 50, 100, 200, 300, 400 ppm of GA₃were used as treatment concentration to study the sprouting ability in off season (April-May) of potato production. Sub title 4: Callus induction and in vitro regeneration of potato Three well established potato cultivar viz. Cardinal, Diamont and Granulla were used for the study.

Achievement/Findings:

The in vitro regeneration and multiplication potentiality was highest in the variety Granulla followed by Cardinal and Diamont. MS + 6% sucrose + 4 mg/L KIN combination of treatment was best for in vitro tuberization among the parameters under study. Off-season sprouting ability and in vitro regeneration potentiality were investigated to establish a rapid multiplication protocol in three potato varieties viz. Diamant, Cardinal and Granulla. It reveal that, sprout initiation time, sprout length and number was directly proportionates to the higher concentration of GA₃ application. Higher concentration (400 ppm) showed better performance for all the parameter under studied. The cultivar Granulla showed best response in callus induction and in virto regeneration under 2,4-D application.
