Effect of Salts on in Vitro Regeneration of Aromatic and Fine Grain Rice.

Abstract/ Executive Summary:

Studies on cultivability with the use of different salt stress the induction and regeneration stages were attempted to enhance regeneration rate of aromatic and fine grain rice in the hope to develop high yielding modern aromatic rice variety. Maximum callus induction was observed in MS medium containing 2.0 mgl-1 2.4-D with lower concentrations of salt, T1, T5 & T9 (2.5 gl^-1). The frequency declines with increased concentration (5.0-10 g/l) of different salts. For callus induction with different salts indicated that NaCl and Na₂So₄ were better than NaHCO₃. In general, the callusing and regeneration ability was found to be inversely proportional to the concentration of different salts.

Objectives:

To determine the effect of salts on in vitro regeneration of aromatic and fine grain rice.

Methodology:

Soil salinity is one of the major environment stresses affecting plant growth adversely. Call and callus cultures are the ideal systems in assessing the physiological effects of salts at the cellular level. Use of stress ingredients such as salt improved regeneration for many rice. Development of regeneration protocol is essential for incorporation of quality traits and developing new varieties by various biotechnological methods. On the other hand the demand of fine and aromatic rice is increasing day by day. Callus induction and regeneration ability of five aromatic and fine grain rice varieties viz; BR5, BRRI dhan28, Hasansarai, Kalizeera and Kataribhog were studied. The seeds were cultured on MS medium containing salts (NaCl, Na₂So₄, and NaHCO₃) at various levels (2.5-10.0 g/l). for regeneration of plantlets callus were transferred to a modified MS medium supplemented with 1 mg/1 NAA, 1 mg/1 kinetin with salts such as NaCl, Na₂So₄, NaHCO₃ (2.5-10.0 g/l).

Achievement/Findings:

Among the varieties BR5 had the highest frequency of callus production (60.81%) followed by Hasansarai (52.5%), Kalizeera (48.16%) ,Kataribhogh (33.66%) and BRRI dhan28 (33.16%). Plant regeneration ability varied significantly among the varieties and affected by media composition. Higher regeneration frequency was observed from medium containing 1.0 mgl^-1 NAA+ 1.0 mgl^-1 Kn with 2.5 hl^-1 NaCl followed by the same treatment of NaHCO₃ and NaSO₄. Among the varieties BR5 had the highest regeneration ability (59%) followed by the Kalizeera (42.41%),Hasansarai (42%), Kataribhogh (42%) and BRRI dhan28 (28%). In general, the callusing and regeneration ability was found to be inversely proportional to the concentration of different salts.
