Diagnosis and Control of Shrimp Diseases

Abstract/ Executive Summary:

Mass larval mortalities have been found in hatcheries due to luminous vibriossis in April-May. Larval mortality was not observed during January to March. A total of 52 water samples from hatchery larval tanks were collected and plated onto agar plates containing thiosulphate citrate bile salt (TCBS) agar medium. Average Vibrios count done in thiosulphate citrate bile salt ‘TCBS’ agar media was between 4.8 x 102 to 3.6 x 103 cfu/ml. The average count of green colonies (produced by luminous bacteria especially Vibrio harveyi) ranged between 8.0 x 101 to 9.7 x 102 cfu/ml. To combat bacterial diseases and larval losses in hatcheries, most operators are accustomed to use a number of antibiotics mainly as precautionary measures without performing antibiotic sensitivity test. Some antibiotics do not work at all against vibrios and rather get resistance. When antibiotics are used to kill certain bacteria, either strains of the pathogens or different bacteria survive because they carry genes for resistance because of indiscriminate antibiotic use. Twelve different antibiotic discs from pharmaceutical company were tested against 52 bacterial isolates collected from different hatcheries. Ciprofloxacin was found to be the best antibiotics against different bacterial isolates followed by Erythromycin and Streptomycin.

Objectives:

To study the prevalence of WSSV in captured broodstock, shrimp post-larvae and cultured shrimp by PCR techniques; and development of suitable management measures for control and prevention of shrimp diseases (WSSV) in hatcheries and farm.

Methodology:

Shrimp (P. monodon) brood stock were collected from fishing trawler (sea) and from different hatcheries. In case of hatchery brood stock, only pleopod was taken from brood stock for PCR analysis without sacrificing the animals. DNA extraction and PCR analysis was done in 'Niribili PCR Lab' at Kalatoli Hatchery Zone, Cox's Bazar, established by the technical guidance of BFRI. To detect WSSV in shrimp post-larvae, samples was collected from different hatcheries. Cultured shrimp were collected live from shrimp culture ponds from Chakaria, Cox's Bazar Sadar, Technaf and Maheskhali.

Achievement/Findings:

Healthy and virus free broodstock will be search to be used to produce virus free post-larvae, key to successful shrimp farming; shrimp reproduction will be increased many fold by stocking healthy and virus free shrimp post-larvae; and knowing the types of antibiotic sensitivity will be helpful to formulate guidelines of uses of antibiotics, which will be also helpful to disease control.
