Morphological and Physiological Characterization of Alternaria Brassicicola Isolates

Abstract/ Executive Summary:

Collection, isolation and characterization of Alternaria brassicicola, a normal pathogen for cabbage, were done to know the details of morphological and physiological characteristics. Samples were collected from different vegetable growing area namely Joydebpur, Sherpur, Commilla, and Ghorasal during 2010-2011 crop seasons. Isolates were examined for colony color, colony texture, and colony diameter. For physiological aspect, growth on PDA and PDA broth was studied under different temperature levels viz 10, 15, 20, 25, 30, and 35^OC

Objectives:

To analysis Alternaria brassicicola isolates

Methodology:

Diseased cabbage samples were collected from different vegetable growing area namely Joydebpur, Sherpur, Comilla, and Ghorasal during 2010-2011 crop seasons. Infected plant parts were collected in polyethylene bag and were taken to the laboratory. Associated fungus was isolated on PDA and pure cultures were done. Single spore culture of fungus was obtained by serial dilution method. All isolates were grown on PDA medium for cultural and morphological variability. The colony diameters were measured on PDA medium poured into 80 mm petri dishes (15 ml/plate) with four replications following the methods of Lilly and Barnett ( 1951) The inoculated plates were incubated at 25±1^OC in incubator for seven days.After incubation, cultures were examined for colony color, colony texture, and colony diameter . For physiological aspect, growth on PDA and PDA both was studied under different temperature levels viz 10, 15, 20, 25, 30, and 35^OC

Achievement/Findings:

A total of 8 isolates of Alternaria brassicicola were collected from surveyed area. Morphological and physiological characteristics of collected isolate were studied. For all isolates, morphological charecteristics of the colony were determined. There were some distinct differences in growth and development of the fungus. Loose and compact textured, different colony color and different colony diameter were recorded from different isolates. One isolate was selected for physiological study which was cultured on PDA and PDA broth was studied at different temperature levels viz. 10, 15, 20, 25, 30, and 35^OC. The highest colony diameter was observed in 25-30^OC temperature
