Studies on Molecular Markers for Varietal Identification of Jute in 2000.

Abstract/ Executive Summary:

The primer OPH-13 had been used in DAMD-PCR for the following jute varieties D-154, BJC-83, BJC-7370, O-4, O-9897, CV-1, CVE-3, CC-45 and genotype incisifolius. This PCR(s) required 2 boosting before the detection of varietal difference. The gel photograph showed the DNA profie after DAMD-PCR with the help of the primer OPH-13. Difference between C.capsularis and C.olitorius was observed and it was four and eight bands respectively. Difference has also been observed within the varieties of C.capsularis and C.olitorius. An exceptional band was observed in CVE-3, BJC-83 & CVL-1. By using more primer (work on which is going on at the moment) definitely more difference will be found out between C.capsularis and C.olitorius and also within varieties.

Objectives:

To identify the varietal differences at molecular level and molecular marker for environmental stress tolerance

Methodology:

In order to determine varietal difference among jute varieties a recently developed method of DNA fingerprinting called (DAMD-PCR (Zhou et al.; 1977) was adapted. This method is reproducible and has been successfully used in rice. The primer OPH-13 had been used in DAMD-PCR for the following jute varieties D-154, BJC-83, BJC-7370, O-4, O-9897, CV-1, CVE-3, CC-45 and genotype incisifolius. This PCR(s) required 2 boosting before the detection of varietal difference. The gel photograph (Fig. Physio-6) showed the DNA profile after DAMD-PCR with the help of the primer OPH-13.
