Production of Cellulase Enzyme from Wild Type and Mutant Strains of Neurospora Crassa

Abstract/ Executive Summary:

N. crassa secretes cellulases to the culture supernatant ,since activity was detected in mycelium cultures of the fungus. The cultures grown on CMC yielding detectable cellulase activities. But cellulase activity of wild type was found to be higher than that of UV ray induced mutated N.crassa . Wild N. crassa showed highest cellulase activity .Mutant – 5 showed nearest and mutant -6 showed lowest activities. UV mutation is a random mutation . So, it may cause some genetic change in the gene that is responsible for cellulase production. All the time UV mutation may not be beneficial.

Objectives:

To produce the cellulase enzyme from wild type and mutant strains of Neurospora crassa .

Methodology:

A wild type and seven mutants of Neurospora crassa was collected from Dhaka University. To investigate the activities of enzymes, wild and mutants of N. crassa were grown on VM liquid medium separately in which the glucose was replaced by carboxy methyl cellulose at 0.5 % concentration. The fungi were incubated at 30 degree centigrade for 3 days in shake culture. The mycelia and other non soluble material in the culture filtrates were separated by centrifugation at 15,000rpm for 5 min. The clear supernatants were used for enzyme assays. For this purpose, standard curves of reducing sugar for CMCase were prepared. Reducing sugars were determined by measuring the absorbance at 540 nm. Hydrolytic activity was calculated by measuring the amount of reducing sugars released from CMC. The amount of reducing sugar was measured by the DNS method using glucose as standards for CMCase activities.

Achievement/Findings:

N,crassa secretes cellulase to the gulture supernatant ,since activity was detected to mycelium cultures of the fungus. But this cellulase activity of wild type was found to be higher than that of UV ray induced mutant N, crassa . The enzymatic assays of this study indicate the presence of a hydrolytic system in N.crassa ,which was able to grow in media containing cellulase as the sole carbon source.
