Effect of Soil Nutrients on Microbial Population in Wetland Rice Cultivation

Abstract/ Executive Summary:

Significantly higher populations of soil bacteria found compared to fungus and actinomycetes. In the complete fertilizer treatment, higher bacterial populations were recorded compared to other treatments. This consequence revealed the missing of one particular major nutrient element either, K, P or N gave similar populations of bacteria. While, the fungus population did not affected by missing nutrients. An increased actinomycetes population was found in missing of P and N nutrient treatments. However, the microbial population may affected by existing rice crop in the field as plant root exudate provide some required nutrients to microbes and every crop cycle added some of root and straw stubbles as organic matter in the missing treatment plots. A soil sampling without crop may generate different scenario of microbial population in missing element experiment. Hence further study needed to find out the effect of missing nutrient on microbial population.

Objectives:

To determine the effect of long term absence of major nutrient elements on total bacteria, fungus and actinomycetes population in wetland rice cultivation system.

Methodology:

The total microbial populations were enumerated from Long trail missing element (-N, -P, -K, control and complete fertilizer treatments) experiment. The invitro studies were conducted at the BRRI soil microbiology laboratory, Gazipur during the period of Boro 2009 to determine the microbial population that affected by the availability of major nutrient elements. The soil samples were collected between four hills and kept in 4^oC temperature until analyses. Exact 1gm of soil sample was taken into 95 ml of sterile distil water and shaken for 15 minutes. A series of 10 fold dilution were prepared up to 10-8 and 0.1ml of each dilution was spread on the media plates. To enumerate total bacterial, fungal and actinomycetes population nutrient agar (NA), potato dextrose agar (PDA) and actinomycetes media plates were used respectively. After 5 days of incubation microbial population was counted following spread plate technique.
