Abstract/ Executive Summary:

The research work was conducted at Biotechnology Institute of Graz Technical University of Austria in 1997 to study between chemical and enzymatic desizing of jute fabrics and its impact. Microorganisms capable of producing alpha-amylase enzyme have been isolated from various decaying starchy materials. Five fungal and three bacterial isolates were purified and selected for further studies. All the five fungal strains produced amylase along with cellulose and hemicelluloses enzymes. The bacterial strains produced mainly amylase enzymes, where starch was the sole source of carbon. The bacterial strain BJB-10053 has been identified as Bacillus amyloliquefaciens, which produces mainly manganese on xylan or galactomannan medium. It produced both amylase and mannanese enzymes.

Objectives:

Production of desizing materials from microbial source suitable for jute yarns and fabrics.

Methodology:

Isolation and purification of amylase producing fungi and bacteria: The microorganisms have been isolated from the collected samples by dilution plate culture method. Some special techniques were followed to isolate hemophilic and alkalophilic strains. After isolation of the bacterial and fungal strains they were purified through repeated and streak culture method. For isolation and purification of the microorganisms, both potato dextrose and starch media were used. Production of amylase under shake culture condition: For shape culture the media used contained the following ingredients: Starch- 10g Yeast extract- 10g Magnesium Sulphat.4 molecule water- 0.04g Magnesium Chloride. 6 molecule water- 0.2g Potassium dihydrogen Phosphate- 1g Dist. Water- 1000cc The initial PH was adjusted with 10% sodium carbonate (w/v) to PH 10-0, if required alkaline otherwise it was adjusted to PH 5.5. Enzyme Assay: Alpha amylase activity was measured at the Biotechnology Institute of Graz Technical University, Austria by courtesy of Prof Dr. Walter Steiner.

Achievement/Findings:

The work has been done on enzyme production by the isolated strains. However, The primary investigation with the selected strains indicates that detailed studies should be carried out in order to find out the optimum fermentation condition for alpha-amylase production.The bacterial strain BJB-10053 has been identified as Bacillus amyloliquefaciens, which produces mainly manganese on xylan or galactomannan medium. It produced both amylase and mannanese enzymes.
