Varietal Improvement and Molecular Characterization of French Bean Genotypes

Abstract/ Executive Summary:

RAPD molecular markers were used to evaluate the genetic diversity in the fourteen varieties of French bean (Phaseolus vulgaris) of three eco-geographical regions of Bangladesh. Out of the 20 random decamer primers used, 6 yielded polymorphic banding patterns. In total, 40 different DNA bands were reproducibly obtained, out of which 28 (70%) were polymorphic. The polymorphisms were scored and used in band-sharing analysis to identify genetic relationships. Cluster analysis based on Jaccard’s similarity coefficient using UPGMA grouped all the 14 genotypes into two major groups. Pair-wise variety comparison of the varieties showed that inter-variety similarity indices for PV004 vs. PV005 (98.51) was higher than all other varietals pairs. On the other hand, inter-cultivar similarity index for BARI 01 vs. PV009 (59.58) cultivar was lower than all other cultivar pairs. Therefore, the inherent simplicity and efficiency of RAPD analyses, coupled with the number of polymorphisms detectable between related genotypes, should facilitate the construction of RAPD-based genetic linkage maps in the context of populations representative of most bean breeding programs.

Objectives:

1. to enrich genetic resources of French bean. 2. to evaluate the performance of collected French bean germplasm 3. to select the promising line/lines which will help to varietal improvement of French bean.

Methodology:

Field experiment was conducted at the Horticultural Research Farm, BSMRAU, Gazipur to investigate the performance of French bean genotypes on the seed yield during the period of November 2008 to March 2009. Eleven French bean genotypes (land races and varieties) collected from different localities of Sylhet region, Chittagong Hill Tracts and released variety of BARI (Bangladesh Agricultural Research Institute) were included in the present investigation. All the cultural management practices has been done as per requirement. The experiment was laid out in a Randomized Complete Block Design with three replications. The whole experimental area was divided into three blocks, representing three replications. Each block was further subdivided into 11 (eleven) unit plots. The eleven different genotypes and varieties of French bean represented as treatments of the experiment were allotted to the 11 unit plots per block. The data of various parameters recorded in the experiment were compiled and statistically analyzed with the help of MSTATC program and the means were separated following Duncan’s Multiple Range Test (DMRT).

Achievement/Findings:

Among the different genotypes studied, PV001 was the highest Yielder compared to others. . For dry seed yield (khaisha purpose) the genotype PV004 was found better. . RAPD analysis revealed high levels of genetic variability, even with the use of limited set of primers. This high level of polymorphism among individuals suggests that RAPD techniques can be useful for French bean for the maintenance of germplasm banks and the efficient selection of parents for breeding.
