Characterization and Documentation of Sugarcane Using Molecular Markers

Abstract/ Executive Summary:

This experiment was conducted to establish varietal identification and determine the variation among the sugarcane varieties/clones through DNA Fingerprinting at BSRI and BAU Lab during the cropping season 2010-2011. This investigation was conducted using 12 sugarcane somaclones of 3 standard varieties such as Isd 37 SC1, Isd 37 SC2, Isd 37 SC3, Isd 37 SC4, Isd 37 SC5, Isd 35 SC1, Isd 35 SC2, Isd 35 SC3, Isd 35 SC4, Isd 35 SC5, Isd 34 SC1, and Isd 34 SC2. An efficient and easy method was adopted for isolation of high quality DNA from meristem cylinder without liquid nitrogen. The amount of recovered DNA was enough for Polymerase Chain Reaction (PCR) amplification and marker studies such as RAPD markers. Electrophorograme of 12 sugarcane somaclones of 3 varieties using one primer. The results of the present investigation revealed that RAPD primer was able to identify the 12 sugarcane somaclones of 3 varieties.

Objectives:

a. To determine the variation among the sugarcane varieties and active germplasm collection of BSRI using molecular markers. b. To establish varietal identification of sugarcane through DNA Fingerprinting. c. To document sugarcane varieties based on molecular markers. d. To develop Marker Assisted Selection (MAS) method for sugarcane.

Methodology:

a. The BSRI-bred varieties, as well as active germplasm collection preferably flowering germplasm those collected from home and abroad were used as experimental materials. b. DNA extraction and purification were followed by the modified mini prep CTAB method. Three individual DNA samples from a cultivar or germplasm in equal volume and concentration were pooled to form the bulked sample. c. RAPDs and SSRs primers were used in PCR amplification. d. Amplified DNA were run in Agarose Gel and Polyacrylamide Gel by electrophoresis to separate allele specific bands. e. Electrophoresis Gel were documented for Fingerprinting using Gel Documentation System,UV-Transilluminator and Digital Camera as and where required. f. Documented Electrophoreses Gel were scored on the basis of the presence or absence of the allele specific bands. Data were analysed to determine the relationships among all cultivars and/or germplasm collection and cluster analysis were carried out using computer software.
