Micropropagation of Sugarcane Varieties for Rapid Multiplication and High Quality Seeds (hqs) Production.

Abstract/ Executive Summary:

This experiment was conducted to optimize variety specific media and explant for micropropagation at BSRI Lab and Field during the cropping season 2010-201. Sugarcane varieties such as Isd 33, Isd 34, Isd 35, Isd 39 and Isd 40 were used to produce high quality seeds (HQS) production. Shoot tips, basal spindle leaf roll transverse sections (TS) and basal spindle leaf segments were used as explants on optimized media. Tops of 6 to 8-month old field-grown sugarcane plants were collected and after removing the outer mature leaves, tops were thoroughly washed with water, sprayed with 70% ethanol and, under aseptic conditions additional outer leaves were removed to isolate immature leaf rolls formed by the innermost 5-6 tightly furled spindle leaves. Shoot tips were isolated carefully with removing tightly furled leaf sheaths with the help of a pair of forceps and scalpel blade. Long spindle (1-2 cm) with axillary shoot buds was cut and placed on medium as shoot tip explant. Firstly, 8-10 cm long basal portion of the leaf roll, starting from the leaf base just above the apical meristem, was exised and placed in liquid MS medium. Each portion was then cut sequentially into 1-2 mm thick TS beginning from the basal end of the roll and placed on the medium. Secondly, Leaf roll was also cut into 4-5 mm long segments and split half way and finally quarterly through longitudinally into four quarters. Then leaf segments were separated individually and placed on the medium. For initial establishment of shoot tips MS (Murashige and Skoog, 1962) medium supplemented with BA (6-Benzyladenine) 2.0 mg/l and Kn (Kinetin) 1.0 mg/l was used. Initial establishment of basal spindle leaf roll TS and basal spindle leaf segments MS medium supplemented with Kn 0.5 mg/l and NAA (α-Naphthaleneacetic acid) 7.5 mg/l was used. Among the three explants shoot tips was more responsive to produce shoots establishment followed by leaf segments and spindle leaf transverse section.

Objectives:

Methodology:

a) The BSRI-bred varieties were included in the investigation. Shoot tip, leaf segments and spindle leaf transverse section were excised and were used as explants for micropropagation. Media were manipulated and optimise for different varieties to get maximum multiplication rate. Micropropagated plantlets were hardened and were planted in the field for performance studies. b) Plantlets produced through shoot tip and meristem culture were supplied to Pathology Division to test against mosaic, red rot and other diseases. c) Genetic stability of micropropagated plants were tested using DNA Fingerprinting.
