Tissue Culture and Utilization of Allied Crops- Stevia, Palm, Date Palm, Sugar Beet and Mushroom

Abstract/ Executive Summary:

This experiment was conducted to find out suitable protocol and sources of explants for micropropagation at BSRI Lab, Field and BAU Lab during the cropping season 2010-2011. Three types of explant viz. young leaves, mature leaves and roots of Arabian and Local date palm were used to induce callus using five different modified MS media viz. MS-1, MS-2, MS-3, MS-4 and MS-5 which were supplemented with different concentrations of 2, 4-D such as 0,1,2,3,4,5,10,20,50 and 100 mg/1 respectively. In all media studied, mature leaves and roots explants did not show any response up to 20 weeks of incubation. In this period, the explants were alive. Only young leaves explant induced to callus formation within eight weeks in dark incubation. Out of five modified media, MS-5 performed better than other media used. In Arabian date palm, the highest percentage (40%) of explant produce callus in MS-5 medium containing 5 mg/1 2,4-D, whereas 25% in Local date palm, in the same concentration of 2,4-D, the lowest percentage (5%) callus produced in MS-1 in Arabian date palm while in MS-2 for Local date palm. Medium containing 5mg/1 of 2, 4-D (MS-5) showed better performance in Arabian date palm compared to other media. Similar results were also found in Local date palm.

Objectives:

a. To identify the suitable sources of explants for micropropagation. b. To find out suitable media for micropropagation. c. To harden plantlets for transplanting. d. To develop low cost method for mushroom seed production through tissue culture. e. To develop tissue culture protocols for improvement and micropropagation of Sugar Beet.

Methodology:

a. The explants, shoot tips, leaf base with petiole and nodal segments of Stevia were collected from 4-6 month old yard grown plants. b. The explants were cut into small pieces (about 1cm long) and then treated with 1% savlon for 5-6 minutes with constant shaking and washed thoroughly with distilled water. c. Then the explants were taken under laminar airflow cabinet. Surface sterilization were done with a 0.1% mercuric chloride solution containing two-three drops of tween-80 for five minutes under aseptic condition. Surface sterilized explants were washed four times with sterilized distilled water. d. Then explants were inoculated aseptically for shooting on MS medium with different concentrations and combinations of growth regulators. Shoots were cultured on rooting media for root production. e. Rooted plantlets were hardened and transplanted in the soil f. Leaves were collected from the mature Stevia plant. After weighing, the leaves were washed with distilled water and oven dried at 100 0C for 48 hours. The dry weight of the leave were recorded. Dry leaves were powdered and used in tea for testing sweetness. g. Shoot tip of Date palm and embryo of Palmyra palm were cultured on MS medium supplemented with different concentrations of cytokinins and auxins for shoot and root production. h. Attempts were taken for Mushroom tissue culture by passing laminar-flow-hood and autoclave to produce Mushroom seeds using tissue culture technique i. Petiol and intact leaves of Sugar beet were used as explants for micropropagation of sugar beet and cultured on MS medium supplemented with different concentration of auxins and cytokinins
