Studies on the Biochemical Basis of Seed Deterioration

Abstract/ Executive Summary:

Results showed that before storing, HC-2 had obtained the highest amount of sugar and it was 264 mg/g of seed. Breeding line CPI-72126/1 had lowest amount of sugar, which was 235mg/g of seed. Among the varieties/line highest amount of sugar was obtained in variety HC-95 and it was 550 mg/g. The other variety HC-2 & breeding line CPI-72126/1 had achieved 312.5 and 380 mg of sugar in per gm of seed respectively. Total protein content of seeds was estimated following the method described by Lowery et.al. in 1951. Drastic decrease of protein was observed in CPI-72126/1 AND IT WAS ONLY 14.50 MG/G OF SEED. The other two varieties (HC-2 & HC-95) had obtained 16.83 and 23.17 ug protein per g of seed respectively.

Objectives:

To find out the seeds deterioration by some biochemical investigation.

Methodology:

Total soluble sugar content was estimated following the method described by Shirlow and Gilchrist (1967). Kenaf varieties HC-2, HC-95 & breeding line CPI-72126/1 were used as test materials.

Achievement/Findings:

Results showed that before storing, HC-2 had obtained the highest amount of sugar and it was 264 mg/g of seed. Breeding line CPI-72126/1 had lowest amount of sugar, which was 235mg/g of seed. After six months of storing in tin container no appreciable difference of sugar content was observed but seeds stored in jute sack showed that sugar content in all varieties/tin increase significantly (at 1% level of probability). Among the varieties/line highest amount of sugar was obtained in variety HC-95 and it was 550 mg/g. The other variety HC-2 & breeding line CPI-72126/1 had achieved 312.5 and 380 mg of sugar in per gm of seed respectively. Total protein content of seeds was estimated following the method described by Lowery et.al. in 1951. Results presented in Table-4 showed that the highest amount of protein (194.70 ug/g seed) was observed in CPI-72126/1 whereas HC-2 and HC-95 had obtained 169.70 and 184.40 ug protein per g of seed respectively. After six months of storage in tin container no appreciable change of protein content of seeds of varieties/line was observed. But after storing in jute sack the level of protein content of seed decreased significantly at 1% level. Drastic decrease of protein was observed in CPI-72126/1 AND IT WAS ONLY 14.50 MG/G OF SEED. The other two varieties (HC-2 & HC-95) had obtained 16.83 and 23.17 ug protein per g of seed respectively.
