Variability Studies of Fusarium Oxysporum F. Sp. Ciceri Causing Chickpea Wilt

Abstract/ Executive Summary:

In vitro studies were conducted on different culture media, temperature, and pH levels on mycelial growth of Fusarium oxysporum F. Sp. ciceri isolates. The fungus isolates showed best growth on Potato dextrose and Richard’s agar media among five culture media that were tried. Growth of Fusarium oxysporum was maximum at 20-30⁰C temperature. The most suitable pH level for growth of fungus was 5.0-6.0.

Objectives:

To characterize some Fusarium isolates of infecting chickpea.

Methodology:

The experiment was conducted to obsarve morphological and physiological veriability of 13 isolates of Fusarium oxysporum f. sp. ciceri collected from major chickpea growing areas of Joydebpur and Ishurdi. Infected plants were collected in polyethylene bag and were taken to the laboratory. The pathogen was isolated using tissue in culture technique. Five to seven days after incubation the fungal culture were studied under stereoscopie and compound microscope for identification of the desired pathogen. After identification the pathogen was purified for further study by single spore isolation technique. All isolates were grown on PDA medium for observation of cultural and morphological variability. The colony diameters were measured on PDA medium poured into 90 mm petridishes (15 ml/plate) with two replications following the methods of Lilly and Barnett (1951). The inoculated plates were incubated at 25±1⁰C in incubator for five days. After incubation, cultures were examined for colony color, texture, colony shape and colony diameter. For physiological aspect, growth on PDA was studied under different temperature levels viz 10, 15, 20, 25, 30 and 35⁰C.

Achievement/Findings:

All the isolates exhibited variation in colony characteristics such as color, shape and texture. Colony colors were Deep pink, Pinkish white, Whitish pink, White. White colour mycelium was common in most of the isolates but other pigmentation such as pink, violet, brown were also prominent. The intensity of colour varied from time to time depending upon the period of incubation. Colony shape was irregular round and slightly round; colony texture as loose, compact and slightly compact. The results of the experiment revealed that, Potato dextrose, Czapek’s Dox agar and Richard agar media were beter for the radial growth of Fusarium oxysporum isolates with a mean maximum growth of 48.75 to 53.75 mm. The effect of temperatures on the mycelial growth of Fusarium oxysporum on PDA is presented. Differences in the growth rate of all the isolates of Fusarium oxysporum were recorded at different temperature levels viz. 10⁰C, 15⁰C, 20, 25, 30 and 35⁰C. At temperature 25⁰C all the isolates showed very good growth and the maximum growth of 90 mm and minimum growth of 63.5mm was recorded. The mycelial colony growth was highest for all isolated at 20-30⁰C temperature. The PH of the culture medium had a significant role on colony growth of Fusarium oxysporum. The fungus grows well on PDA media with a wide range of PH 4.5 to 7.0 in this study. For this study, two isolate were selected. The highest colony growth was observed at 5.0-6.0 PH. The maximum growth was 60.25 to 63.55 mm and minimum growth was 45.50 mm recorded.
