Morphological and Physiological Characterization of Alternaria Brassicicola Isolates

Abstract/ Executive Summary:

Collection, isolation and characterization of Alternaria brassicicola, a common pathogen for cabbage, were done to know the details of morphological and physiological characteristics. Isolates were examined for colony color, colony texture, and colony diameter.The effect of culture media, temperature and pH on growth of Alternaria brassicicola was studied and found that Corn meal auger, Potato dextrose, and Czapek’s Dox agar media were the best for the growth of the fungus. The highest colony diameter was observed in 25-30^OC temperature and pH 5-6 with maximum growth at 25^oC.

Objectives:

To study the effect of culture media, temperature and pH on the growth of Alternaria brassicicola.

Methodology:

Diseased cabbage samples were collected from different vegetable growing area namely Joydebpur, Sherpur, Narsingdi and Jessore during 2011-2012 crop seasons. Infected plant parts were collected in polyethylene bag and were taken to the laboratory. Associated fungus was isolated on PDA and preparation of pure cultures was done. Single spore culture of fungus was obtained by serial dilution method. All isolates were grown on PDA medium for cultural and morphological variability. The colony diameters were measured on PDA medium poured into 90 mm petri dishes (15 ml/plate) with four replications following the methods of Lilly and Barnett (1951). The inoculated plates were incubated at 25±1OC in incubator for seven days. After incubation, cultures were examined for colony color, colony texture, and colony diameter. For physiological aspect, growth on PDA was studied under different temperature levels viz 10, 15, 20, 25, 30, 35^OC and Potato Dextos Bron (PDB) broth was studied at 25 ^OC.

Achievement/Findings:

A total of 9 isolates of Alternaria brassicicola were collected from surveyed area. Morphological and physiological characteristics of these isolates were done in the laboratory. For all isolates, morphological characteristics of the colony were determined. There were some distinct differences in the growth and development of the fungus. Loose and compact textured, different colony color and different colony diameter were recorded from different isolates. The results of the experiment revealed that, Corn meal auger, Potato dextrose, and Czapek’s Dox agar media were the best for the radial growth of Alternaria brassicicola isolates with a average maximum growth of 32.25 to 36.75 mm.
