Morphological Characteristics of Some Isolates of Sclerotium Rolfsii and Fusarium Spp

Abstract/ Executive Summary:

Twenty two isolates of Sclerotium rolfsii and 13 isolates of Fusarium spp were tested to identify their morphological characteristics. For this purpose pathogens were grown on PDA and PD broth at 25⁰C temperature and various radial growth, sclerotia number and mycelial dry weight were recorded.

Objectives:

To compare growth behavior and morphological similarities and differences among the collected isolates of Sclerotium rolfsii and Fusarium Spp .

Methodology:

Twenty two isolates of sclerotium rolfsii and 13 isolates of Fusarium spp were collected from different locations and different hosts. Collected isolates were purified and preserved on PDA for morphological characteristics. For measurement of radial growth, 3 days old cultures were grown on PDA at 25⁰C and data were recorded after 6 days. Number of sclerotia was counted per cm². For determination of mycelial dry weight PD broth was prepared and 3 days old mycelial blocks were cultured on the broth. After seven days mecelial dry weight was measured.

Achievement/Findings:

After 6 days of culture on PDA, different isolates of Sclerotium rolfsii showed different mycelial growth rate and number of sclerotia. Mycelial growth rate ranged from 1.47mm to 9.55mm where as the number of sclerotia varied from 1.0 to 4.0 per cm2. On the other hand, radial growth of of Fusarium sp. was ranged from 6.0mm to 9.20 mm after 6 days of inoculation . Mycelial dry weight varied from 0.07gm to 0.29gm
